The Pennsylvania Railroad main line between Pittsburgh and Chicago was the main railroad through Fort Wayne, using its own station. At Adams, 5 miles east of Fort Wayne, a PRR's Fort Wayne Branch diverged south to Richmond IN. And at Junction, 1 mile west of Fort Wayne, PRR's Grand Rapids Division diverged north, terminating in Mackinaw City.

Fort Wayne was located on the Wabash main line between Detroit and St. Louis, with Wabash using its own station. At New Haven, 6 miles east of Fort Wayne, a branch line diverged east to Toledo.

Fort Wayne was located on the Nickel Plate main line between Buffalo and Chicago, with NKP using its own station. Nickel Plate also had the New Castle Division, extending south.

New York Central's Fort Wayne Branch extended north to Hillsdale and Jackson MI, with trains using NYC's Cass Street Depot.
